Cyanobacteria are the primary producers of geosmin and 2-methylisoborneol (2-MIB), which contribute to undesirable taste and odor in drinking water and fish globally Recently, there has been an increase in outbreaks of cyanobacterial blooms in various water bodies.
The Tri An and Dau Tieng Reservoirs in Vietnam have increasingly raised concerns regarding taste and odor issues.
The present study identified the off-flavor compounds geosmin and 2-MIB in field samples and 50 isolated strains of planktonic cyanobacteria from Dau Tieng and Tri An Reservoirs For the first time, these compounds were screened using headspace solid-phase micro-extraction and gas chromatography–mass spectrometry (HPME GC–MS) The ability to produce off-flavor compounds was linked to the presence of two molecular markers: geoA and mib Results indicated that mean concentrations of geosmin and 2-MIB ranged from 13.3 to 9.6 ng/L and 4.9 to 101.7 ng/L, respectively, in field samples, with a total of five strains producing geosmin.
The internship report by instructor Do Thi Ngoc Nhi highlights the identification of five geosmin-producing strains with a maximum concentration of 272.9 ng/g wet weight (ww) and nine strains producing 2-MIB, with the highest concentration reaching 362.1 ng/g ww These strains belong to two genera of cyanobacteria, Dolichospermum and Oscillatoria, with four of the geosmin producers also capable of producing 2-MIB The maximum likelihood (ML) tree analysis of 16S rRNA demonstrated a high diversity among the isolated strains, representing various morpho-types and genotypes Additionally, BLAST searches revealed the genetic sequences of the 16S rRNA obtained during the research.
