device[1], using evacuated-vial-eluted, disposable and sterile cartridges to increase the concentration of 99m Tc/ 188 Re of commercial 99m Tc/ 188 Re generator eluates, was developed for the effective utilisation of Technetium in Technegas generator-based lung perfusion ( 100-250 mCi/mL) and other SPECT (20-30 mCi/mL) imaging.
Trang 5A self-shielded radioisotope concentrator device (Fig 2) was created based on
a newly developed sorbent / concentrator column which selectively retains
is then eluted from the concentrator column with 1.0 mL saline into an evacuated vial through a millipore filter and ready for injection The liquid flow effected by the vacuum of the evacuated vial and the septum-needle connection–based fluid communication design of the device was optimised to make elution process effective, simple, sterile and radiation safe This design is
to ensure the vacuum-operated and multi-elution use in the form of
generator (ANSTO, Australia) of 110 GBq activity eluted with 10 mL saline was chosen to test our radioisotope concentrator device (Fig.1c in the upper right corner) Gamma-ray spectrometry and Capintec dose-calibrator were used for radiometry assays.
Trang 6The 99m Tc eluate was concentrated more than 10-fold with a 99m Tc recovery yield of > 85% using this radioisotope concentrator device 5 repeated elutions were successfully performed with each cartridge So, each cartridge can be effectively used for one week
in the hospital environment for radiopharmaceutical formulation The useful lifetime of the 99m Tc generator was significantly extended depending on the activity of the generator as shown in below table The 99 Mo impurity detectable in the 99m Tc solution eluted from the Gentech generator (Fig 1b) was totally eliminated by this radioisotope concentrator device and ultra-pure, concentrated 99m Tc-pertechnetate solution was achieved The concentrated 99m Tc solution is well suited to labeling in-vivo kits and to loading the crucibles of Technegas generators for V/Q SPECT imaging.
Conclusion:
We conclude that the radioisotope concentrator device functioned well and is robust in operation This is a user-friendly and effective device for extending the operational lifetime of generators by providing a concentrated 99m Tc/ 188 Re solution which can be used in Technegas V/Q SPECT lung perfusion and other SPECT imaging This device will,
to some extent, mitigate the global 99m Tc crisis The material is a work-in-progress and patent-pending.
